Analysis of the genetic determinism of the mineral concentration of cow's milk

The composition in minerals of cow's milk plays an important role in its nutritional value and its cheese-making abilities. By combining high-throughput phenotyping with middle infrared spectrometry (MIR), large-scale genotyping, association and annotation analyses, 50 regions of the genome were detected that affect the main mineral concentrations, identifying the genes implicated and candidate genetic variants (usually involved in regulation). The genes SLC37A1 and ANKH explain the most significant QTL.

The main mineral concentrations (calcium,magnesium, phosphorus, potassium, sodium and citrate) can be predicted on a large scale using middle infrared spectrometry (MIR). This high-throughput phenotyping leads to genetic studies on trait determinism. This study used MIR predictions from a million samples from 26 876 Montbeliard cows. The traits studied had a high heritability  (0.50-0.58), except for sodium (0.32). From the statistically reconstructed (imputed) genome sequence for 19 586 genotyped cows, a genome-wide association study (GWAS) detected 50 regions of the genome affecting these traits, identifying the genes implicated and proposing candidate genetic variants, for the most part in the non-coding sequences. Analyses (in silico) show 877 regulating candidate variants that alter transcription factor (TF) binding sites or situated in non-coding RNA. The genes SLC37A1 and ANKH, coding for the proteins implicated in ion transport, explain the most significant QTL. This study shows the group of genes and variants affecting the mineral content of milk.
